We're getting a 500 error when trying to communicate from a remote to
the redirector.

We've tried configuring IIS to run 32 bit applications via:
cscript.exe adsutil.vbs set W3SVC/AppPools/Enable32BitAppOnWin64 true

...thinking that IIS was having a problem executing the 32 bit dll.

IIS is configured as per the steps in the whitepaper: "Running
MobiLink Redirector on a Microsoft Windows Server 2003 Enterprise
Edition Server" :

We were able to successfully communicate using a Win 2003 32 bit
redirector. However, not with the 64 bit.

As far as I know, this should work regardless of the OS bitness. Investigating further.

One thing you need to make sure of on the 64-bit machine is that all the Redirector supporting files
are 32-bit. The Redirector can only load 32-bit files, so if you have a mixed bitness of files in
your Redirector setup, this could cause the failure.
