Hello!

Is it possible to upgrade from SQL 2008 to SQL 2008 R2, or is it necessary
to uninstall the previous version first?

In general you can, but it depends from what edition/architecture you

Same Platform, same Edition => Windows Server 2008 R2 64bit, SQL Server 2008
Enterprise 64bit => SQL Server 2008 R2 Enterprise 64bit

How to upgrade? Start setup.exe and let the installer do the upgrade?? ...or
do I have to do some [required] steps before upgrading? Any advice?

I have not performed any upgrade, but I would expect that you just
insert the disc, and select the option to upgrade. But if you navigate
from the link that I gave you, you should be able to find a topic
about upgrading. Look at the TOC to the left and there a couple of
topics that can be of interest.
