We create lot of DTS in our server. Is it possible to make the DTS read only?

Read only?

Do you mean

1. Can view a package's contents but not execute
2. Can execute a package but not view the contents?

You may want to look at applying passwords to the packages (Owner and User)
to see if that is what you need.
